BrainBits® Papain Protocol For Cell Dissociation
Enclosed is one glass vial containing 6 mg sterile papain and one 15 mL
centrifuge tube containing 5 mL of Hibernate® E without Calcium
(BrainBits® HE-Ca 5 ml) for improved cell yield during dissociation.
Preparations (Room Temperature in a Sterile Hood):
1. The cell dissociation solution is prepared by adding 3 mL of the
supplied HE-Ca to the 6 mg of sterile papain (BrainBits® PAP 6 mg) for
a final working concentration of 2 mg/ml papain.
2. Incubate for 10 minutes at 30ºC to dissolve.
3. Once dissolved, add 2 mL of cell dissociation solution to freshly
dissected BrainBits® CNS tissue and incubate for 10 min at 30oC.
Gently swirl every 5 minutes.
4. Disperse and plate cells as per BrainBits® Primary Neuron Culture
